I would also add:
Who is my audience?
What are their primary concerns?
What would potential objections be to my message?"
How do I overcome potential objections?
How do I get my message fixed in their memory?

I agree with Parissa, "Who is my audience?" is the first question I ask:
- What is their level of understanding of the subject?
- How much time do they have to spend with me?

Our questions are surprising alike with just nuanced differences.

1) What is the goal of the presentation.
2) WIFM - Not the radio station analyzing the audience for the hook. What Is In It For the Me. You must tell your audience what is in it for them if you are going to keep them engaged, you must understand and view your slides through the WIFM lens to keep them engaged.
3) What pain does this presentation cure and how do I express or get the audience to feel that pain in the opening. Without pain, there is no change.

You need to consider your audience's expectations, needs, interests, and motivations, as well as their demographics: age, gender, income level, education, etc.

Next, you need to consider your purpose for the presentation: what do you want to acheive? Do you want people to understand an issue, agree with your recommendation, or buy something?

The last question encompasses all the techniques of good communication: given this audience and this goal, what strategies do I need to employ to acheive my goal? Some audiences relate more strongly to the speaker than others. Some situations call for strong emotional appeals while other audienes and situations call for more logical appeals.

Great question and one of my favorite topics! When I prepare for a presentation I ask myself:

1) What is my objective for this presentation - what do I want the audience to do with the information I am presenting.

2) What challenges are the audience faced with and how can I personalize the experience so my content resonates with them.

3) What is the benefits I am teaching or bringing to the group to entice them to particpate and listen to my presentation to know this I must know who my audience is and what thier challenges are.

Who is my audience and how can I show them the WIIFM and the connection to business strategy quickly, to obtain their buy-in?
What are my goals and how can my objectives ensure I meet them and create measurable outcomes?
What is the best medium for delivering this content and in what time frame? (E-learning, IL formal facilitation, self-study, SME, etc...)
How should I follow up after the course to ensure the couse is everything attendees need and that key course content is relevant on the job?
I know that's four, but they're all so very important!
