Typically the A: drive is a floppy (ie: 3.5").
Do you have it installed?

If the device has failed, it may not show as avalable any longer.

The drive may be defective, or a cable may have come loose. Just to check it's not your OS (I'm supposing this to be a flavo(u)r of Windows) that is failing to detect the drive (while it is in perfect working order), download, burn and boot from a Linux live-CD to check this.
BTW: most BIOSes allow you to boot from the floppy (and/or check its presence during the boot process). Enable (either one of) these options and see if the drive actually spins up (and/or the activity LED lights up).
Just my 2 €cents...

Does device manager see it? Anything flagged with a yellow triangle and asterisk?
If it's not a USB floppy then there aren't any drivers to go wrong under windows - so most likely a hardware problem. If it's a desktop, crack the case with the power off and reseat the floppy ribbon cable at both ends, and reseat the power connector. If all else fails, swap the floppy out - they're standard and this should only take a minute.
